The Effect of Serum on the PEG-Based Crosslinker-Induced Spheroid Formation of Pancreatic -Cell

چکیده
A polymeric crosslinker poly(ethylene glycol) derivative with dioleyl groups as hydrophobic group at both ends was developed for promoting cellular spheroid formation. Our approach to bridge cells was based on the crosslinking of cell membrane using a crosslinker via the hydrophobic interaction. Using the crosslinker, spheroid formation of pancreatic -cell line RIN in a round bottom 96-well plate was evaluated, especially in effect of serum on spheroid formation. In the presence or absence of serum, the size of prepared spheroid was found to decrease with increasing culture time and with increasing crosslinker concentration. However, the medium without serum proved to be a favorable condition for promoting cell aggregation because in this case spheroid with smaller size could be obtained. It was clarified that spheroid formation and insulin secretion of the spheroid prepared by the crosslinker were enhanced, especially in the medium

Comparison of radiosensitizing effect of Resveratrol on monolayer and spheroid culture of DU145 prostatic cell line
Background: Radiotherapy is an established therapeutic modality for prostate cancer. Resveratrol, a natural antioxidant, has been shown to inhibit carcinogenesis and to block the process of tumor initiation and progression. No data is available on the response of cellular spheroid to Reseveratol. In this study we have examined the effect of Resveratol on the radiation response of human p...
متن کاملThe Effect of Wild Type P53 Gene Transfer on Growth Properties and Tumorigenicity of PANC-1 Tumor Cell Line
The p53 protein function is essential for the maintenance of the nontumorigenic cell phenotype. Pancreatic tumor cells show a very high frequency of p53 mutation. To determine if restoration of wild type p53 function can be used to eliminate the tumorigenic phenotype in these cells, pancreatic tumor cell lines, PANC-1 and HTB80, differing in p53 status were stably transfected with exogenous wil...
متن کاملUrtica Dioica Distillate Regenerates Pancreatic Beta Cells in Streptozotocin-Induced Diabetic Rats
Background: Urtica dioica is known as an anti-hyperglycemic plant. Urtica dioica distillate (UD) is a traditional Iranian drink, locally known as “aragh gazaneh”. In spite of its widespread consumption in Iran, according to traditional Iranian medicine, there is no scientific report on the usefulness of UD for diabetic patients. This survey was designed to evaluate its protective effects for th...
متن کاملThe effect of leaf extract P.harmala on streptozotocin-induced apoptosis in pancreatic tissue of rats
Abstract Background and Purpose: Decreased beta-cell mass has a role in the pathogenesis of human type 2 diabetes and in rodent diabetes models. The study of plant extracts is important as they possibly inhibit the reduction of beta cell mass and the islets of Langerhans. The effect of the methanolic extract of P. harmala leaves on pancreatic tissue was investigated in this research. Method: A ...
متن کاملThe role of Rad51 protein in radioresistance of spheroid model of DU145 prostate carcinoma cell line
Background: Rad51 is a protein with critical role in double strand break repair. Down-regulation of this protein has a significant effect in radiosensitivity of some cell lines like prostate carcinoma. Compared to monolayer cell culture model, the spheroids are more resistant to radiation. The aim of the current study was to determine the Rad51 protein level in DU145 spheroids, and monol...
